چکیده

Materials and Methods. Materials. Protease-inhibitor cocktail tablets and sequencing grade trypsin were obtained from Roche. N-octyl-glucoside was from Calbiochem, and tissue culture reagents were from Life Technologies. Tetracyclin-free foetal bovine sera (FBS) was from Perbio, and other tissue culture reagents were from Biowhittaker. Precast 4–12% and 10% polyacrylamide Bis-Tris gels were obtained from Invitrogen; P81 phosphocellulose paper was from Whatman; Phenformin from Sigma and AICA riboside from TorontoXX. [γP]ATP, glutathione-Sepharose, Protein G-Sepharose were purchased from Amersham Biosciences. All peptides were synthesised by Dr Graham Bloomberg at the University of Bristol.

LKB1 is a master kinase that activates 13 kinases of the AMPK subfamily, including MARK/PAR-1.

We recently demonstrated that the LKB1 tumour suppressor kinase, in complex with the pseudokinase STRAD and the scaffolding protein MO25, phosphorylates and activates AMP-activated protein kinase (AMPK). A total of 12 human kinases (NUAK1, NUAK2, BRSK1, BRSK2, QIK, QSK, SIK, MARK1, MARK2, MARK3, MARK4 and MELK) are related to AMPK. Here we demonstrate that LKB1 can phosphorylate the T-loop of a...

The Par-1/MARK family of protein kinases: from polarity to metabolism.

The Par-1 protein kinases are conserved from yeast to man and belong to a subfamily of kinases that includes the energy sensor and metabolic regulator, AMPK. Par-1 is regulated by LKB1 and atypical PKC and has been shown in multiple organisms and cell types to be critical for regulation of cellular polarity. Recent studies using knockout mice have revealed several surprising physiological funct...

P20: The Role of Protein Kinases in Memory

When an experience is encrypted into a long-lasting memory, it is believed that specific sets of neurons in the brain of the animal undergo changes including the strengthening of preexisting synapses and the growth and maintenance of new synaptic connections. These activity-dependent synaptic changes appear to require the coordination of a variety of cellular processes in spatially separated ce...

Investigation the Mechanism of Interaction between Inhibitor ALISERTIB with Protein Kinase A and B Using Modeling, Docking and Molecular Dynamics Simulation

The high level of conservation in ATP-binding sites of protein kinases increasingly demandsthe quest to find selective inhibitors with little cross reactivity. Kinase kinases are a recently discovered group of Kinases found to be involved in several mitotic events. These proteins represent attractive targets for cancer therapy with several small molecule inhibitors undergoing different ph...

Cellular SRC kinases and dsRNA dependent protein kinase (PKR) play key role in intracellular viral (CVB3) replication

SRC kinases and PKR are intracellular protein kinases, which play key roles in intracellular viral replication. In this research, the effect of SRC kinase inhibition and PKR activation and inhibition on replication of coxsakievirus (CVB3), an entrovirus of the family picornaviridae – causative agents of fatal myocarditis, was studied. Vero and Hela cells were cultured and infected with CVB3 in ...
